Synthesis and characterization of polysaccharide-oligonucleotide conjugate

Abstract

We have reported that SPG can from a complex with oligodeoxynucleotide (ODN) attaching (<A20) tail at 3′ end. However, SPG cannot form the complex with ODN attaching short tail (<dAso) due to reduction of thermodynamic stability. In this study, we examined whether ODN-conjugated SPG can form an intramolecular complex. dA20 was introduced to reducing terminal of SPG via diaminoalkanes or diaminoPEGs. The conjugates were purified by gel-filtration. CD spectrum of the conjugate indicated a formation of intramolecular complex.
