Quantitation of tetrodotoxin in human plasma with mixed solvent and combined cartridges

Abstract

In order to improve the recovery of tetrodotoxin (TTX) in human plasma, our extraction and purification procedures were further evaluated. For the effective extraction of TTX, the use of mixed solvents, methanol-acetonitrile, with the ratio of about 8:2 was recommended. For the purification of TTX by the first cartridge, 3 kinds of cartridges, Sep-Pak C 18, Sep-Pak tC 18 and PS-2 were equally useful when complete removal of protein and lipids was carried out at the extraction step. In the purification of C 9-base by the second cartridge, PS-2 gave best recovery with one-hour holding time. The established method should be useful for the trace analysis of TTX in human plasma.
