Peptide array for the comprehensive analysis of intracellular signals aiming to drug screening

Abstract

New peptide array, on which many substrate peptides for protein kinases are immobilized, was developed. If the array was treated with isolated protein kinases or cell lysate, each peptide was phosphorylated dependent on their properties, after the optimized immobilization and masking procedures. Phosphorylation in each spot was detected by fluorescence on glass slide or 2 dimensional SPR on gold chip using anti-phospho antibody. In the glass array using Cy-3 labeled antibody for the detection, drug-specific profiles of peptide phosphorylation were obtained. Using this method, new assay system for the drug screening or fuctional analysis of unknown gene may be possible.
