N2 - The increase of errors and the decrease in number of learned mice were significantly ameliorated by Crocus sativus ethanol extracts (CSE) in SD test. The LTP-blocking effect of ethanol was significantly improved by oral-, intravenous-, and intracerebroventricular- administration of CSE, respectively. When a single oral administration of crocin was given 10 min before the ethanol treatment the number of successful mice increased at a dose of 200 mg/kg. Crocin of 50 mg/kg ameliorated the blocking effect of ethanol on the LTP at 84% compared to the control. Crocetin gentiobiose glucose esteralso antagonized the blocking effect of ethanol on the LTP dose-dependently indicating about a half of crocin. Crocetin di-glucose ester did not remove the inhibitory effect of ethanol on the LTP. It is concluded that two gentiobiose moieties are necessary for the appearance of pharmacological activity of crocin in the central nervous system.
