Discrimination of bark borer-resistant clones of Cryptomeria japonica using RAPD markers

Abstract

We have developed useful RAPD (random amplified polymorphic DNA) markers to discriminate among 38 clones of Sugi (Cryptomeria japonica D. Don) that have resistance to attack by Cryptomeria Bark Borer (Semanotus japonicus L.), which clones were selected in 1998 and 2001. A primary screening was performed for 100 arbitrary detainer primers, and 18 primer that generated 25 clear and polymorphic fragments were selected. When RAPD analysis was repeated two times, all selected RAPD markers confirmed the reproducibility. As a result, 34 of 38 resistant clones were discriminated by using these RAPD markers. The others share the same profile. They would be so related since they are from the same area and /or the same local race. Moreover, we evaluated the discrimination powers of these 18 primers. Three of the 18 primers had high discriminatory powers of more than 0.5.
