D-RECS (4) a novel gene carrier responding to intracellular protease signal

    We recently proposed an artificial gene regulation systems responding to the intracellular protease signal, such as Caspase signal, which is the key signal of the cell death. For this purpose, we designed polymer-peptide conjugate for a gene carrier. This polymer interacts with DNA through an electrostatic interaction and suppressed the gene-transcription. On the other hand, in the presence of active Caspase, this complex is disintegrated and the gene transcription is recovered. In this report, we applied this system to the cell free system and cultured cell, and succeeded in regulating the delivered gene expression responding to the intracellular protease signal.
